Overview: Anacazole is a third-generation aromatase inhibitor, commodity named Ruiyu, Reininger, chemically named 2,2'---5-(1H-1, 2,4-Sanjuan-2 1-base-methyl)-1, 3-subbenzene double (2-methyl propylene), is a third generation selective nonsteroidal aromatase inhibitor developed by AstraZeneca for clinical use in the treatment of advanced breast cancer in postmenopausal women who are ineffective after treatment with TAM and other anti-estrogen therapy, First-line treatment for advanced breast cancer in postmenopausal women and early breast cancer in postmenopausal women. The pharmacological effect of anatroclone acts on the last step of the estrogen synthesis response, inhibiting aromatic enzyme activity and thus reducing estrogen levels in the body fluid circulation. Anacoo or 10mg/d can inhibit aromatase activity by 96.7% from the baseline, estrogen, estradiol and esoxycin activity by≥ 86.5% ≥ 83.5% and ≥ 93.5%, respectively. There was generally no difference in the levels of estradiol inhibition of 0.5 or 1.0 mg/d, but the level of estradiol after 24h was lower than the average test level, indicating that the minimum dose required for estradiol to maximize inhibition was 1 mg. Even after a few days of interruption, estrogen is continuously suppressed. This product has no effect on the serum concentration of gonadotropins and adrenal corticosteroids, nor does it impair the corticosteroid response stimulated by adrenal corticosteroids (ACTH).
